Telomerase is a ribonucleoproteinase necessary for the replication of chromosome ends in most eukaryotic organisms. It is active in progenitor cells and cancer cells, and is inactive or very low in normal somatic cells. The main activity of the catalytic component of the teloprotease holoenzyme complex is to act as a reverse transcriptase, by duplicating the template sequence in the RNA component of the enzyme, adding simple repetitive sequences to the end of the chromosome, thereby extending the telomere, using the 6-nucleotide end The granule repeat unit 5'-TTAGGG-3' catalyzes the RNA-dependent extension of the 3'-chromosome ends. The catalytic cycle includes primer binding, primer extension, and product release after reaching the template boundary, or displacement of new products, and then further extension to be more active on substrates containing 2 or 3 telomeric repeats. Telomerase activity is regulated by many factors, including telomerase complex-related proteins, chaperones, and polypeptide modifications. The telomerase reverse transcriptase (TERT) gene is highly expressed in stem cells and is silenced during differentiation. Cancer cells can be immortalized by activating TERT to maintain telomere length and telomerase activity, which is a key step in tumorigenesis. TERT gene promoter mutations are mainly manifested as the genetic changes of malignant tumor-specific somatic cells. There are characteristic telomere lengthening in many tumors such as glioma, which is the key cause of tumor occurrence and development. There are two main mutations in the TERT promoter region that are common in gliomas: 1 295 228C>T and 1 295 250 C>T, which are called C228T and C250T, respectively, and are common in glioblastoma and oligodendroglioma .
