PIK3CA gene encodes PIK3CA protein, PI3Kp110a, which is the p110 catalytic subunit of class I phosphatidylinositol-3 kinases (PI3Ks) and belongs to the phosphatidylinositol-3 kinase (PI3Ks) family. The PI3Ks family participates in a variety of signal pathways and regulates cell functions. Under normal circumstances, the lipid products produced by its activation can be used as second messengers to bind and activate multiple target proteins in cells, forming a signal cascade complex, and ultimately regulating cell proliferation, differentiation, survival and migration. The PI3K-Akt signaling pathway in the PI3Ks family is dysregulated in human tumors. The gain or loss of functions caused by mutations in certain components of this pathway can cause cell transformation. This pathway regulates tumor cell proliferation and survival, and is closely related to tumor invasion and metastasis. PI3Kp110a encoded by PIK3CA is the only gene in the PI3K family that has been found to cause somatic mutations and cause cancer.

PIK3CA is a proto-oncogene located on chromosome 3, with a total of 20 exons. About 80% of PIK3CA mutations occur in two hotspots, Helical and Kinase. A large number of data indicate that the mutation hotspots of PIK3CA are concentrated on exon 9 and exon 20, of which the three most common The mutations are H1047R in exon 20, and E542K and E545K in exon 9.

These mutations can all lead to enhanced lipid kinase activity of PI3K. It has been found that there are PIK3CA gene mutations in breast cancer, non-small cell lung cancer and other cancers. PIK3CA gene mutations cause the continuous activation of the PI3K/Akt signaling pathway. PI3K as a downstream signaling molecule of EGFR is activated, leading to tumor cells to EGFR-TKI, etc. Drug resistance.
